Having been a flight attendant for over 17 years, as well as a flight attendant supervisor, I can tell you I did not have a special certification. Once hired, I had to complete 5 weeks of training that covered everything from service, emergencies, medical situations, evacuations, etc.
Perhaps the DOT Training referenced means that a new hire has to have shown as completed training that contains specific topics, areas of expertise to do the job, which would be accounted for in the 5 week training.
I would encourage you to check out specific airlines' career sections of their websites for more detailed information and requirements.

Roman, I just did a quick look around the internet and could not find anything indicating the supv must obtain the FAA medical certification.
The FAA does have requirements for all flight attendants to have certification as to having completed the required training. It is in section 814 of this law. It is not just for supervisors. The need for training standards is pretty common in a lot of professions. Police, lawyers, etc. A LOT can go wrong on a plane. I have met some remarkable flight crew members who were able to stay calm and take control. We need well-trained flight crew!
